NaV1.7 is a voltage-gated sodium channel primarily expressed in peripheral sensory neurons; acting as a "threshold channel," it lowers the threshold for action potential firing by amplifying sub-threshold depolarization, thereby modulating neuronal excitability and driving the generation of pain signals. Inhibiting NaV1.7 reduces the excitability of sensory neurons, resulting in an analgesic effect.
