CSNK1G3/CK1g3

Completing the gamma subfamily, CK1γ3 (gene CSNK1G3) is a membrane-associated, lipid-modified CK1 isoform expressed prominently in brain and kidney. It phosphorylates the Wnt co-receptor LRP6 to potentiate canonical Wnt signaling and has been detected at the centrosome and in trafficking vesicles. In cultured cells CK1γ3 overexpression alters microtubule organization and cell adhesion, and it has been proposed to influence axon growth in neurons. Relative to CK1γ1 and CK1γ2, CK1γ3 is the least studied gamma isoform, with no firmly established disease phenotype, so recombinant CK1γ3 is mainly used to dissect isoform redundancy in membrane-proximal CK1 signaling.
