Zinquin
Based on 1 Customer Validation
Zinquin is a fluorescent zinc ion (Zn2+) indicator. Zinquin can form a fluorescent complex with Zn2+, which is used to study zinc distribution in cells and tissues, zinc-related physiological processes, and changes in intracellular zinc homeostasis (Ex/Em = 368/490 nm).

Biological Activity
Description
In Vitro
1. Stock solution preparation
1.1 Solvent: anhydrous MDSO.
1.2 Recommended concentration: approximately 25 mM.
2. Working solution preparation
2.1 Diluent: physiological saline (124 mM NaCl, 2.5 mM KCl, 2 mM CaCl2, 2 mM MgCl2, 23 mM NaHCO3, 10 mM glucose, bubbled with 95/5% O2/CO2 gas mixture).
2.2 Working concentration: 25 μM Zinquin.
2.3 Note: Adjust the working solution concentration as needed; prepare fresh before use.
3. Staining procedure
3.1 Sample type: rat hippocampal or neocortical slices (400 μm) prepared using a McIlwain tissue chopper; before staining, keep the slices in physiological saline in an interface holding chamber at room temperature.
3.2 Incubation conditions: continuously perfuse the slices with physiological saline containing 25 μM Zinquin at room temperature.
4. Controls
4.1 In liposome experiments, the membrane-permeable Zn2+ chelator TPEN quenches Zinquin fluorescence; no signal is generated after pre-incubation with TPEN.
5. Detection and analysis
5.1 Instrument type: fluorescence microscope.
5.2 Wavelength settings: excitation wavelength 360 nm; 400 nm dichroic mirror; emission filter 510/80 nm.
5.3 Result analysis: Zinquin fluorescence is expected to be visible in the dentate gyrus hilus and mossy fiber projection areas, with less labeling of cell bodies; individual mossy fiber terminals are distinguishable at high magnification.
