Ac-NKFD-AMC

Ac-NKFD-AMC is a synthetic fluorescent peptide substrate for detecting non-apoptotic caspase activity. Ac-NKFD-AMC is cleaved by non-canonical activated caspases produced during monocyte-macrophage differentiation and anti-inflammatory macrophage polarization, and is not cleaved by recombinant apoptotic caspase-8 or caspase-3. Ac-NKFD-AMC can specifically distinguish non-apoptotic caspase enzymatic activity under macrophage polarization states.

Ac-NKFD-AMC detects non-apoptotic caspase activity in human primary monocyte-derived M0 and IL-4-polarized M2 macrophages, which depends on CTSB and is absent in M1 macrophages and apoptotic monocytes^[1].
Ac-NKFD-AMC is not cleaved by recombinant active caspase-8 and caspase-3 in this cell-free enzymatic reaction^[1].
