ADEP 19
ADEP 19 is a ClpP1P2 dysregulator with antibacterial activity against Mycobacterium tuberculosis and various Gram-positive bacteria. ADEP 19 binds to the ClpP2 subunit of Mycobacterium tuberculosis, disrupting ClpP1P2-ClpX/ClpC1 chaperone interactions. ADEP 19 inhibits ClpC1P1P2-mediated proteolytic activity (IC50 = 1.2 μM) while upregulating the ATP hydrolysis activity of ClpXP1P2. ADEP 19 exhibits synergistic effects with Bedaquiline (HY-14881) against Mycobacterium tuberculosis. ADEP 19 can be used in research related to bacterial infections, such as tuberculosis.

Biological Activity
Description
In Vitro
ADEP 19 (4-20 μg/mL; 4 days after 14 days of hypoxia) reduces the viability signal of non-replicating hypoxic Mtb H37Rv[1].
ADEP 19 transiently increases and subsequently decreases intracellular ATP levels in Mtb H37Rv[1].
ADEP 19 (7 days) in combination with Bedaquiline (HY-14881) produces a strong synergistic antibacterial effect against Mycobacterium tuberculosis (ΣFICI = 0.375); in combination with Rifampicin (HY-B0272), Isoniazid (HY-B0329), and Streptomycin (HY-B1906), no synergistic effect exists[1].
ADEP 19 (24 hours) exhibits potent antibacterial activity against various Gram-positive bacteria, including Staphylococcus aureus (MIC = 0.125 μg/mL), Enterococcus faecium (MIC < 0.008 μg/mL), Streptococcus pneumoniae (MIC < 0.008 μg/mL)[1]
ADEP 19 (4-20 μg/mL; 0-14 days) alone completely kills Mtb H37Rv after 14 days and enhances the bactericidal effect of BDQ in time-kill assays[1].
ADEP 19 (10 μM; 24 h) accumulates intracellularly in Mtb H37Rv, reaching 80% in the cell uptake assay[1].
