AX-202
Based on 1 Customer Validation
AX-202 is a humanized anti-S100A4 monoclonal antibody that neutralizes S100A4. AX-202 blocks downstream profibrotic and proinflammatory effects. AX-202 exerts antifibrotic effects in skin fibrosis. AX-202 induces regression of pre-established skin fibrosis. AX-202 modulates anti-inflammatory and antifibrotic transcriptional pathways. AX-202 inhibits STAT3 phosphorylation. AX-202 induces AMPK and CASQ1 expression. AX-202 modulates SSc-related transcriptional signatures. AX-202 reduces tenascin-C, a DAMP and endogenous TLR4 ligand. AX-202 can be used for the research of systemic sclerosis.

Animal Model:C57BL/6J (male) mice injected with Bleomycin[1]
-
Dosage:8 mg/kg; 16 mg/kg; 24 mg/kg
-
Administration:i.p. every third day for 3 weeks; i.v. once weekly for 3 weeks
-
Result:Across all regimens, dermal thickness, myofibroblast count, and hydroxyproline content were reduced versus bleomycin alone.
With 16 mg/kg i.p. every third day and 24 mg/kg i.v. once weekly, dermal thickness and myofibroblast counts fell below those of mice given bleomycin for 3 weeks then NaCl for 3 weeks; dermal thickness was reduced by more than 60% versus persistent fibrosis.
pSTAT3-positive cells and tenascin-C staining intensity were reduced and AMPK- and CASQ1-positive cells were increased in bleomycin-challenged skin.
RNA sequencing identified 506 regulated DEGs (277 up-regulated, 229 down-regulated); 53 of 258 DEGs shared between bleomycin-induced fibrosis and SSc (20.5%) were regulated.

Flow cytometric analysis of 1X106 Jurkat cells labeling AX-202 (HY-P991363, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. AF 488-conjugated AffiniPure Goat Anti-Human IgG H&L (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG4 kappa (HY-P99003, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black). -
Immobilized N-hFChuS100A4 can bind AX-202. The EC50 for this effect is 55.38 ng/mL.
