ZEN-3694
Based on 1 publication(s) in Google Scholar
ZEN-3694 (BET-IN-19) is an orally active BET inhibitor. ZEN-3694 regulates the function of BET proteins, thereby downregulating pathways involved in cell cycle regulation, ER signaling, AR signaling, AR splice variants, MYC, GR, cell growth, inflammation, and cellular immune responses. ZEN-3694 inhibits proliferation, induces apoptosis, reverses the upregulation of CDK6 and CCND1, restores cell cycle arrest, and inhibits CDK6 via miR-34a-5p. ZEN-3694 can be used in research related to HER2 breast cancer and metastatic castration-resistant prostate cancer.

ZEN-3694 acts synergistically with abemaciclib to inhibit the proliferation of MCF-7, PALBO-R MCF-7 and ABEMA-R MCF-7 cells, with combination index (CI) values ranging from 0.07 to 0.36[1].
ZEN-3694 (administered for 5 days) potently reduces the viability of palbociclib-resistant MCF7-R100 and T47D-R100 ER+ breast cancer cells cultured under palbociclib treatment, with a nanomolar-level efficacy; compared with parental MCF7 cells, the resistant cells show higher sensitivity to this drug[3].
ZEN-3694 (administered for 14 days) inhibits long-term colony formation of palbociclib-resistant MCF7-R100 and T47D-R100 ER+ breast cancer cells cultured in palbociclib, with nanomolar potency, and the resistant cells exhibit higher sensitivity than the parental MCF7 cells[3].
ZEN-3694 (1 μM) downregulates the elevated CDK6 protein levels in PALBO-R and ABEMA-R MCF-7 cells, and reduces ERα protein levels in MCF-7, PALBO-R MCF-7 and ABEMA-R MCF-7 cells[1].
ZEN-3694 alters the expression of over 11,000 genes and inhibits key oncogenic pathways (including estrogen-mediated S-phase entry, STAT3 and VEGF signaling pathways) in MCF-7, PALBO-R MCF-7 and ABEMA-R MCF-7 cells[1].
Combination treatment with ZEN-3694 and abemaciclib induces high levels of apoptosis in wild-type and CDK4/6 inhibitor-resistant ER+ breast cancer cells after 7 days of treatment (75.2% in MCF-7 cells, 60.2% in PALBO-R MCF-7 cells, and 50.4% in ABEMA-R MCF-7 cells)[1].
ZEN-3694 induces dose-dependent decreases in the protein expression of CDK6, cyclin D1, cyclin E2, phosphorylated Rb, ERα and c-MYC in palbociclib-resistant MCF7-R100 ER+ breast cancer cells[3].

In Vivo:
Select the appropriate dissolution method based on your experimental animal and administration route.
- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (7.50 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (7.50 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
