2-Hydroxy-dATP diphosphatase is a Nudix sanitizing enzyme found in eukaryotes, bacteria and archaea; it hydrolyzes oxidized purine triphosphates such as 2-hydroxy-dATP to prevent their misincorporation into DNA, and also recognizes 8-oxo-dGTP and 8-oxo-dATP with lower efficiency (cf. EC 3.6.1.55). It hydrolyzes 2-hydroxy-dATP into 2-hydroxy-dAMP and diphosphate. In cells it sanitizes oxidized dATP. It features a Nudix fold.
