Aconitate hydratase (aconitase) is an iron-sulfur enzyme containing a [4Fe-4S] cluster that interconverts citrate, cis-aconitate and isocitrate, with an equilibrium mixture of about 91% citrate, 6% isocitrate and 3% aconitate. cis-Aconitate is (Z)-prop-1-ene-1,2,3-tricarboxylate. It catalyzes both dehydration (citrate to cis-aconitate) and rehydration (to isocitrate), occupying the TCA-cycle and glyoxylate-cycle branch point. The cytosolic aconitase/IRP1 also acts as an iron-regulatory RNA-binding protein. Structurally it is a [4Fe-4S] aconitase.
