Acrocylindropepsin is an aspartic endopeptidase from the imperfect fungus Acrocylindrium sp., with a very low pH optimum of 2.0 on casein, belonging to peptidase family A1. It prefers hydrophobic residues at P1 and P1', and on the insulin B chain behaves like pepsin A but additionally cleaves Leu6-/-Cys(SO3H), Glu21-/-Arg and Asn3-/-Gln, though not Gln4-His. In the fungus it digests extracellular proteins under strongly acidic conditions. It features a pepsin-like aspartic-protease fold.
