Adenain is a cysteine endopeptidase from adenoviruses, the type example of peptidase family C5, with a protein fold unlike that of any other known peptidase. Its activity is greatly stimulated when an 11-residue peptide from the adenovirus capsid protein pre-VI binds at a site separate from the active site. It cleaves adenovirus and host-cell proteins at two consensus sites, -Yaa-Xaa-Gly-Gly-/-Xaa- and -Yaa-Xaa-Gly-Xaa-/-Gly-, where Yaa is Met, Ile or Leu and Xaa is any amino acid. In the viral life cycle it matures the capsid to render it infectious. It features a beta-barrel fold with a catalytic Cys-His-Glu triad activated by the viral pVI peptide cofactor.
