Adenosylhomocysteinase is an NAD+-containing C-S bond hydrolase that hydrolyses S-adenosyl-L-homocysteine into L-homocysteine and adenosine; each subunit tightly binds one NAD+, which transiently oxidises the 5'-deoxyadenosine residue at C-3', labilising the thioether bond in a mechanism analogous to that of inositol-3-phosphate synthase (EC 5.5.1.4). By clearing the feedback inhibitor S-adenosylhomocysteine it is the only known route to remove the by-product of all cellular methyltransferases and is essential for transmethylation, adenosine metabolism and detoxification of the ethionine toxin. It is distributed across Bacteria, Eukaryota and Archaea and adopts a tetrameric NAD+-bound aldehyde-dehydrogenase-like fold.
