Alpha,alpha-trehalose synthase is a trehalose-biosynthetic enzyme that requires Mg2+. It reversibly transfers glucose from NDP-alpha-D-glucose onto D-glucose, releasing NDP and forming alpha,alpha-trehalose. In the reverse direction it is specific for alpha,alpha-trehalose and cannot use aryl glucosides, maltose, sucrose, lactose or cellobiose. The Rubrobacter xylanophilus and Pyrococcus horikoshii enzymes use ADP-, UDP- and GDP-alpha-D-glucose equally; Thermococcus litoralis prefers ADP- and Thermoproteus tenax prefers UDP-glucose. It occurs in Bacteria and Archaea.
