Alternative-complement-pathway C3/C5 convertase (C3bBb/C3bBb3b) is a bimolecular complex of complement fragment Bb with C3b or cobra-venom factor, Bb containing the active site and formed when factor D cleaves proenzyme factor B; cleavage of C5 requires additional C3b to bind C5, and C3bBb is stabilised in plasma by properdin (factor P); factor B is in family S1; it cleaves the Arg-Ser bond in the C3 alpha chain to C3a and C3b and an Arg bond in the C5 alpha chain to C5a and C5b. By amplifying complement activation it drives alternative-pathway defence. It appears in viruses and cellular organisms.
