EC Tree > 3 Hydrolases > 3.4 Acting on peptide bonds (peptidases) > 3.4.21 Serine endopeptidases > 3.4.21.111 aqualysin 1
Aqualysin 1 is an alkaline serine peptidase from the extreme thermophile Thermus aquaticus in family S8A, with three substrate-binding subsites S1, S2 and S3, preferred residues at S1 being Ala and Phe, at S2 Ala and norleucine, and at S3 Phe and Ile (specificities similar to peptidase K EC 3.4.21.64 and subtilisin BPN' EC 3.4.21.62); it shows broad specificity on the insulin B chain and hydrolyses elastin substrates such as succinyl-(Ala)n-p-nitroanilide (n=1,2,3) and some peptide esters, with low specificity toward esters of amino acids with small hydrophobic or aromatic P1 residues. By digesting proteins at high temperature it supports thermophilic bacterial nutrient acquisition. It is taxonomically restricted to Thermus.
