Archaeosine synthase is a tRNA-modifying amidinotransferase. In Euryarchaeota it is ArcS; in Crenarchaeota lacking ArcS it is carried by a glutaminase-domain-containing 7-cyano-7-deazaguanine synthase (EC 6.3.4.20) or a preQ1 synthase (EC 1.7.1.13) homologue. It transfers the amidino group from L-glutamine onto the 7-cyano-7-carbaguanine15 of tRNA, hydrolyzing glutamine to glutamate and water and forming archaeine15. The Methanocaldococcus jannaschii enzyme also uses arginine and ammonium. It occurs in Archaea, Eukaryota and Bacteria.
