Atrolysin C is a 24-kDa hemorrhagic zinc metalloendopeptidase from the western diamondback rattlesnake (Crotalus atrox) venom that digests type IV collagen, existing as two forms c and d, in peptidase family M12; hemorrhagic toxin-2 of C. ruber ruber has the same Mr and specificity. Phosphoramidon inhibits in the 0.1 mM range. It cleaves His5-/-Leu, His10-/-Leu, Ala14-/-Leu, Tyr16-/-Leu and Gly23-/-Phe in the insulin B chain, and with small-molecule substrates prefers a hydrophobic residue at P2' and a small residue such as Ala or Gly at P1. In venom it damages basement membranes. It features an M12-family zinc catalytic domain.
