Microbial collagenase is a zinc metalloendopeptidase from Clostridium histolyticum, with six forms acting on native collagen: class I (alpha, 68 kDa; beta, 115 kDa; gamma, 79 kDa) and class II (delta, 100 kDa; epsilon, 110 kDa; zeta, 125 kDa), immunologically cross-reactive but with complementary specificities. Variants have been purified from Bacillus cereus, Empedobacter collagenolyticum, Pseudomonas marinoglutinosa and related species. The enzymes also act as peptidyl-tripeptidases. They digest native collagen in the triple-helical region at -/-Gly bonds, preferring Gly at P3 and P1', Pro and Ala at P2 and P2', and hydroxyproline, Ala or Arg at P3'. In clostridial infections they destroy host connective tissue. They feature a zinc catalytic domain specialized for helical collagen.
