Biotin synthase

Biotin synthase is the radical-SAM sulfur-insertion enzyme. It binds [4Fe-4S] and [2Fe-2S] clusters and consumes two AdoMet molecules per cycle: the first abstracts H from C9 to make 4,5-dethiobiotin secobiotin, the second abstracts C6 methylene H and the radical closes the thiophane ring via an intramolecular C-S bond. The [2Fe-2S] cluster is the sacrificed sulfur donor (single turnover in vitro, reconstituted in vivo by Isc/Suf). Expected in Bacteria, Eukaryota and Archaea.
