C-mannosyltransferase is a glycosyltransferase that transfers a D-mannosyl residue from dolichyl D-mannosyl phosphate onto a tryptophan residue in protein acceptors, forming a C-C glycosidic bond rather than the more common O- or N-linkage. It occurs in viruses and cellular organisms and carries out protein C-mannosylation, a rare post-translational modification found on the first tryptophan of the WXXW motif in thrombospondin type-1 repeats and other secreted and membrane proteins.
