Calicivirin is a cysteine endopeptidase encoded by noroviruses and other caliciviruses, which are a major cause of epidemic acute viral gastroenteritis. The nonstructural proteins of these viruses are produced by proteolytic cleavage of a large precursor polyprotein performed by a protease incorporated into the polyprotein; cleavage sites are defined by sequence and structural features. It prefers cleavage when P1 is occupied by Glu-/- and P1' by Gly-/-. In the viral life cycle it matures the replicase polyprotein. It features a chymotrypsin-like cysteine protease fold.
