Calpain-3 is a Ca2+-dependent cysteine protease found in skeletal muscle and genetically linked to limb-girdle muscular dystrophy type 2A. It is activated by autoproteolytic cleavage of insertion sequence 1 (IS1), which allows substrates and inhibitors access to the active site; substrates include the enzyme itself and connectin/titin. It belongs to peptidase family C2 and has broad endopeptidase activity and specificity. In muscle it contributes to sarcomere remodeling and repair. It features a calpain catalytic core with distinctive IS1 and IS2 insertions.
