Calpain is a Ca2+-dependent intracellular cysteine protease that reversibly modulates protein function through limited, site-specific cleavage rather than bulk degradation. It participates in cytoskeletal remodeling, cell-cycle progression, apoptosis signaling and membrane-cytoskeleton attachment. In organisms it contributes to muscle plasticity, cell motility and Ca2+-triggered signaling. It features a cysteine-protease catalytic domain, a Ca2+-binding EF-hand regulatory domain and a calmodulin-like small subunit.
