Caspase-1
Definition:
EC Tree > 3 Hydrolases > 3.4 Acting on peptide bonds (peptidases) > 3.4.22 Cysteine endopeptidases > 3.4.22.36 caspase-1
Caspase-1 is an inflammatory caspase from mammalian monocytes, part of the inflammatory caspase family that also includes human caspase-4 (EC 3.4.22.57) and caspase-5 (EC 3.4.22.58) and murine caspase-11, -12, -13 and -14. It contains a caspase-recruitment domain (CARD) in its N-terminal prodomain that participates in procaspase activation, and cleaves pro-interleukin-1beta. It has a strict requirement for Asp at P1 and a preferred cleavage sequence Tyr-Val-Ala-Asp-/-. In organisms it drives pyroptosis and maturation of inflammatory cytokines. It features a catalytic p20/p10 heterodimer with a cysteine-histidine catalytic dyad.
