Caspase-2
Definition:
EC Tree > 3 Hydrolases > 3.4 Acting on peptide bonds (peptidases) > 3.4.22 Cysteine endopeptidases > 3.4.22.55 caspase-2
Caspase-2 is an initiator caspase, together with caspase-8 (EC 3.4.22.61), caspase-9 (EC 3.4.22.62) and caspase-10 (EC 3.4.22.63). It contains a caspase-recruitment domain (CARD) in its N-terminal prodomain that participates in procaspase activation. Two antagonistic forms exist: caspase-2L induces programmed cell death, while caspase-2S suppresses it. It has a strict requirement for Asp at P1, with Asp316 essential for activity, and a preferred cleavage sequence Val-Asp-Val-Ala-Asp-/-. In cells it participates in stress-induced apoptosis and the DNA-damage response. It features a CARD domain fused to a p19/p10 catalytic heterodimer.
