Caspase-5
Definition:
Caspase-5 is an inflammatory caspase, part of the family that also includes human caspase-1 (EC 3.4.22.36) and caspase-4 (EC 3.4.22.57) and murine caspase-11, -12, -13 and -14. It contains a CARD in its N-terminal prodomain for procaspase activation, and can cleave itself and the p30 fragment of gasdermin D. It has a strict requirement for Asp at P1, with a preferred cleavage sequence Tyr-Val-Ala-Asp-/- and also cleaving at Asp-Glu-Val-Asp-/-. In cells it acts as a cytosolic lipopolysaccharide sensor and drives pyroptosis in concert with caspase-4. It features a CARD-p20-p10 architecture.
