Cathepsin H is a lysosomal cysteine peptidase present in mammalian cell lysosomes, belonging to peptidase family C1 (papain family). It hydrolyzes proteins acting both as an aminopeptidase, notably cleaving Arg-/- bonds, and as an endopeptidase. It contributes to lysosomal bulk protein turnover and to the generation of antigenic peptides for MHC presentation. In cells it supports lysosomal homeostasis and tissue remodeling. It features a papain-like fold plus an integral minigonadotropin-like octapeptide that confers aminopeptidase activity.
