Cathepsin K is a lysosomal cysteine endopeptidase in peptidase family C1 (papain family), prominently expressed in mammalian osteoclasts and believed to play a central role in bone resorption. It has broad proteolytic activity; with small-molecule substrates and inhibitors, the major determinant of specificity is P2, preferably Leu, Met > Phe, and not Arg. In the osteoclast it degrades type I collagen and other bone matrix proteins. It is a major drug target for osteoporosis. It features a papain-like fold with an unusual collagenase-specific occluding loop.
