Cathepsin S

Definition:

EC Tree > 3 Hydrolases > 3.4 Acting on peptide bonds (peptidases) > 3.4.22 Cysteine endopeptidases > 3.4.22.27 cathepsin S
Cathepsin S is a lysosomal cysteine endopeptidase in peptidase family C1 (papain family), unusual among such enzymes for remaining stable at neutral pH. Its specificity resembles cathepsin L, but it shows much less activity on Z-Phe-Arg-NHMec and more activity on the Z-Val-Val-Arg-NHMec compound. In antigen-presenting cells it mediates invariant chain degradation and antigenic peptide generation for MHC II. In tissues it contributes to extracellular matrix remodeling, angiogenesis and autoimmune disease. It features a papain-like fold adapted for endosomal/lysosomal and extracellular activity.
