Cholate-CoA ligase requires Mg2+. The mammalian membrane-bound enzyme catalyzes the first step in the conjugation of bile acids with amino acids, converting bile acids to their acyl-CoA thioesters; chenodeoxycholate, deoxycholate, lithocholate and trihydroxycoprostanoate also act as substrates. The soluble bacterial enzyme participates in anaerobic bile-acid 7α-dehydroxylation. Structurally it is a bile-acid CoA ligase (BAAT).
