Classical-complement-pathway C3/C5 convertase (C4b2a/C4b2a3b) is a serine-protease complex of complement fragments C4b, C2a and C2b, with C2a containing the active site and C2b the C4b-binding site, formed when C1s cleaves proenzyme C2 (C2a is in peptidase family S1; includes former EC 3.4.21.44); cleavage of C5 requires C3b to bind C5 and render it susceptible to the C4b2a complex; it selectively cleaves the Arg-Ser bond in the C3 alpha chain to C3a and C3b, and an Arg bond in the C5 alpha chain to C5a and C5b. By generating anaphylatoxins and opsonins it drives classical-pathway inflammation and lysis. It appears in viruses and cellular organisms.
