Mycolysin is a zinc metalloendopeptidase characterized from Streptomyces griseus, S. naraensis and S. cacaoi, the type example of peptidase family M5. Its specificity resembles thermolysin but it is much more sensitive to inhibition by sulfanylacetyl-Phe-Leu, and shows little structural similarity to other bacterial metalloendopeptidases. It preferentially cleaves bonds with hydrophobic residues at P1'. In streptomycetes it digests extracellular proteins. It features an M5-family zinc catalytic fold.
