Complement subcomponent C1s is a classical-pathway serine protease in family S1 activated from proenzyme C1s by C1r; it cleaves the Arg-Ala bond in C4 to form C4a and C4b and the Lys(or Arg)-Lys bond in C2 to form C2a and C2b, assembling the classical-pathway C3 convertase (C4b2a). By cleaving C4 and C2 it drives classical complement activation. It appears in viruses and cellular organisms.
