Cyclic GMP-AMP synthase (cGAS) is the mammalian cytosolic DNA sensor. It condenses ATP and GTP into cyclic Gp(2'-5')Ap(3'-5') (via a pppG(2'-5')A intermediate), releasing two diphosphate; the cGAMP second messenger binds STING and triggers type-I interferon and cytokine production. It occurs in viruses and cellular organisms.
