Deoxyhypusine synthase

Definition:

EC Tree > 2 Transferases > 2.5 Transferring alkyl or aryl groups, other than methyl groups > 2.5.1 Transferring alkyl or aryl groups, other than methyl groups (only sub-subclass identified to date) > 2.5.1.46 deoxyhypusine synthase
Deoxyhypusine synthase is the eukaryotic eIF5A-activating enzyme. In four steps it NAD+-dependently dehydrogenates spermidine, transfers the 4-aminobutylidene group to an active-site Lys (Lys329 in human), then onto a specific lysine of the eIF5A precursor, and reduces the imine with NADH, forming a deoxyhypusine residue on eIF5A and releasing propane-1,3-diamine. The plant enzyme can use homospermidine for spermidine. Hypusine is completed by EC 1.14.99.29. It occurs in Eukaryota, Archaea and Bacteria.
