Dihydrofolate synthase

Definition:

EC Tree > 6 Ligases > 6.3 Forming carbon-nitrogen bonds > 6.3.2 Acid—amino-acid ligases (peptide synthases) > 6.3.2.12 dihydrofolate synthase
Dihydrofolate synthase ( folylpolyglutamate synthase, FPGS) adds the first glutamate to 7,8-dihydropteroate: ATP plus 7,8-dihydropteroate plus L-glutamate yields ADP plus phosphate plus 7,8-dihydropteroylglutamate. In some bacteria a single protein carries both this activity and EC 6.3.2.17 tetrahydrofolate synthase, together producing polyglutamylated H4PteGlun; in most eukaryotes the two activities reside on separate proteins. Structurally it is an FPGS ATP-grasp enzyme.
