E1 NEDD8-activating enzyme
Definition:
EC Tree > 6 Ligases > 6.2 Forming carbon-sulfur bonds > 6.2.1 Acid-thiol ligases > 6.2.1.64 E1 NEDD8-activating enzyme
E1 NEDD8-activating enzyme activates the ubiquitin-like protein NEDD8 in an ATP-dependent manner: ATP plus NEDD8 plus the E1 L-cysteine yields AMP plus diphosphate plus E1-S-(NEDD8)-yl-L-cysteine. NEDD8 conjugation (neddylation) activates cullin-RING ligase (CRL) E3 complexes, which cannot bind substrate directly and instead assemble on cullin scaffolds with substrate-recognition modules. Like ubiquitin, NEDD8 is first adenylated and then transferred to the catalytic cysteine of E1. Structurally it is a heterodimeric NEDD8 E1 (APP-BP1/UBA3).
