IgA-specific metalloendopeptidase is a 190-kDa zinc metalloprotease found in pathogenic Streptococcus species such as S. sanguis and S. pneumoniae, the type example of peptidase family M26. It cleaves the Pro-/-Thr bond in the hinge region of the human IgA heavy chain, which is distinct from the IgA-specific serine endopeptidase (EC 3.4.21.72). In pathogens it cleaves mucosal IgA to enable immune evasion and colonization. It features a zinc metalloprotease fold secreted into the mucosal niche.
