Lysostaphin is a zinc-dependent 25-kDa endopeptidase from Staphylococcus simulans, the type example of peptidase family M23. It lyses S. aureus cells by acting on the cross-bridges of the cell wall, hydrolyzing the -Gly-/-Gly- bond in the pentaglycine inter-peptide link joining staphylococcal cell-wall peptidoglycans. In the bacterium it kills competing staphylococci. It features an M23-family zinc catalytic fold specialized for the pentaglycine bridge.
