Furin (PACE) is a constitutive secretory-pathway proprotein convertase in family S8 structurally and functionally similar to kexin, activated by Ca2+, containing Cys near active-site His and inhibited by p-mercuribenzoate; at least three related mammalian PCs (PC2, PC3, PC4) have somewhat different specificities; it releases mature proteins from proproteins by cleavage of Arg-Xaa-Yaa-Arg bonds where Xaa is any amino acid and Yaa is Arg or Lys, releasing albumin, complement C3 and von Willebrand factor from precursors. By processing growth factors, receptors and viral glycoproteins it is a key secretory-pathway convertase and a viral-entry cofactor.
