Deuterolysin is a ~20-kDa zinc metalloendopeptidase found in Penicillium roqueforti, P. caseicolum, Aspergillus sojae and A. oryzae, the type example of peptidase family M35, formerly in EC 3.4.24.4. It digests various proteins at pH optimum 5, acts strongly on protamine and histones, and is insensitive to phosphoramidon. It preferentially cleaves bonds with hydrophobic residues at P1', and also Asn3-/-Gln and Gly8-/-Ser in the insulin B chain. In fungi it digests nuclear and basic proteins. It features an M35-family zinc catalytic fold.
