Hef endonuclease is a structure-specific archaeal/eukaryotic endonuclease that cleaves the DNA strand at the 5' side of nicked or flapped positions in duplex DNA. By processing stalled replication forks and branched DNA intermediates it contributes to DNA replication restart and homologous recombination in archaea and eukaryotes. It appears in viruses and cellular organisms and contains an XPF-like nuclease domain.
