Hepacivirin (HCV NS3/4A protease) is a hepatitis-C-virus serine protease in family S29 encoded by the HCV genome, contributing to maturation of precursor polyproteins and greatly activated by binding the 54-residue NS4A cofactor also derived from the viral polyprotein; crystallographic structure shows a chymotrypsin-like fold; it hydrolyses four peptide bonds in the viral precursor polyprotein, commonly with Asp or Glu in P6, Cys or Thr in P1 and Ser or Ala in P1'. By processing the HCV polyprotein it drives viral replication and is a major antiviral drug target.
