Histolysain is a cysteine endopeptidase from the protozoan Entamoeba histolytica, in peptidase family C1 (papain family). It hydrolyzes proteins including basement membrane collagen and azocasein, preferentially cleaving Arg-Arg-/- bonds in small-molecule substrates such as Z-Arg-Arg-NHMec. In the parasite it contributes to tissue invasion, host protein degradation and evasion of the immune response. It features papain-like catalytic isoforms secreted to degrade the host extracellular matrix.
