Hyaluronan synthase is a membrane-embedded glycosyltransferase that alternately adds beta-D-glucuronic acid and N-acetyl-beta-D-glucosamine from UDP-alpha-D-glucuronate and UDP-N-acetyl-alpha-D-glucosamine onto the growing nascent hyaluronan chain, releasing UDP. The Streptococcus group A and C enzyme requires Mg2+; it adds GlcNAc when the non-reducing end is GlcA and GlcA when the end is GlcNAc, and is highly specific for UDP-GlcNAc and UDP-GlcA. Similar enzymes occur in many organisms. It occurs in viruses and cellular organisms and synthesizes the vertebrate extracellular matrix glycosaminoglycan hyaluronan.
