Isocitrate lyase is an aldehyde-lyase of the glyoxylate cycle. It cleaves (1R,2S)-1-hydroxypropane-1,2,3-tricarboxylate (isocitrate) into succinate and glyoxylate, bypassing the two decarboxylation steps of the TCA cycle so that cells grow on C2 compounds such as acetate or ethanol. It is absent in vertebrates (which lack the glyoxylate cycle) but present in bacteria, fungi, plants and some invertebrates. Structurally it is a TIM-barrel Mg2+-dependent lyase of the isocitrate-lyase superfamily.
