Fragilysin is a zinc metalloendopeptidase in peptidase family M10 from Bacteroides fragilis, thought to cause diarrhea in animals and humans. It hydrolyzes extracellular matrix proteins, disrupts tight junctions of intestinal epithelial cells, and degrades intracellular cytoskeletal proteins such as actin and myosin. It shows broad proteolytic specificity, cleaving -Gly-/-Leu-, -Met-/-Leu-, -Phe-/-Leu-, -Cys-/-Leu- and Leu-/-Gly bonds. In the pathogen it destroys the intestinal barrier. It features an M10-family zinc catalytic domain.
