Kumamolysin is a bacterial pepstatin-insensitive carboxyl proteinase isolated from Bacillus sp. MN-32 and several Burkholderia spp., exhibiting a Ser278/Glu78/Asp82 catalytic triad and the type example of family S53; it specifically hydrolyses the Leu15-Tyr16 bond in oxidised insulin B chain, additionally cleaving Phe25-Tyr26 at a much lower rate, and cleaves Phe-L-(p-nitrophenylalanine) in synthetic peptides, preferring peptides with Ala or Pro at P2 and charged residues Glu or Arg at P2', with no cleavage of Asp-Pro-Ala-Lys-Phe-pNF-Arg-Leu. By digesting proteins at acidic pH it supports bacterial extracellular proteolysis. It is distributed across Bacteria and Eukaryota.
