Lactocepin (Lactococcus cell-envelope proteinase, PrtP) is a cell-envelope-associated subtilisin-like serine protease in family S8 attached to Lactococcus lactis by a C-terminal membrane anchor, responsible for casein hydrolysis in milk and provision of peptides essential for growth, important in cheesemaking and lactic-casein production for rapid growth to high cell density with adequate lactic-acid production; specificity differences between lactocepins from starter strains may partly explain different cheese flavours; it has very broad endopeptidase activity with noted subsite preferences (large hydrophobic residues at P1 and P4, Pro at P2), best known on caseins but also hydrolysing haemoglobin and oxidised insulin B chain. By digesting milk casein it supplies peptides for dairy fermentation.
