Lipoyl synthase

Lipoyl synthase is the radical-SAM enzyme that installs the lipoyl cofactor. It carries two [4Fe-4S] clusters and, using two AdoMet molecules, abstracts H from C6 and C8 of the octanoyl chain on the lipoyl-domain lysine; radicals interact with sulfur atoms from the auxiliary cluster, forming two C-S bonds on (R)-lipoate. The auxiliary cluster is sacrificed and regenerated by Isc/Suf. Lipoylation activates pyruvate/2-oxoglutarate/branched-chain dehydrogenase E2 domains and glycine-cleavage H protein; an alternative pathway uses EC 6.3.1.20 lipoate ligase. It occurs in viruses and cellular organisms.
